Abstract

Blue pigments are the rarest in nature and the most difficult to obtain, so the industry has been applying synthetic colorants such as brilliant blue (E133), indigo carmine (E132), and patent blue V (E131) in food products. The blue flowers of Clitoria ternatea L. have been identified as a potential source of alternative natural dyes, due to the high color stability and rare hue achieved by the presence of acylated anthocyanins of the ternatin class. In this sense, the present work focused on the characterization of the anthocyanin profile and the evaluation of the bioactive properties of blue petals of C. ternatea. The anthocyanin composition of the hydroethanolic extract was characterized by HPLC-DAD-ESI/MS. The antioxidant activity was evaluated by three different methodologies (OxHLIA, reducing power, and DPPH), the anti-inflammatory activity was evaluated in rat macrophage cells (RAW 264.7), the cytotoxicity in human tumor cells by sulforhodamine B method (MCF-7, NCI-H460, AGS, and CaCo-2), the hepatotoxicity in non-tumor cells (PLP2 and VERO), and the antimicrobial activity was tested against twelve food pathogenic microorganisms. The phenolic characterization revealed the presence of seventeen anthocyanin derivatives, compounds that justify the intense blue hue of the floral extract. Regarding bioactivities, the anti-inflammatory activity (IC50 94±1 µg/mL) was the most promising. The absence of toxicity was proven (PLP2 and VERO, IC50>400 µg/mL), the best cytotoxicity results were 297±18 µg/mL (CaCo) and 341±3 µg/mL (MCF-7), the antioxidant activity was notable in the reducing power (EC50 0.78±0.03 mg/mL), while the fungicidal activity was achieved in concentrations ≤ 1mg/mL for six fungi studied and E. coli (MIC 0.5 mg/mL) was the most sensitive bacteria. According to the results, it can be stated that the floral extract of C. ternatea presented significant bioactivities, provided by its anthocyanin profile, characterizing it as a functional matrix of great importance for the industry.
