IDENTIFICATION OF PHENOLIC COMPOUNDS FROM PITANGA SEEDS (Eugenia uniflora L.)

Resumo

Eugenia uniflora L. is a Brazilian native fruit, has an elevated amount of anthocyanins, phenolics compounds and its seeds can be used as by-product. The aim of this study was to identify the phenolic compounds from purple and red pitanga seeds. Seeds were dried and ground. The moisture was also determined. The extraction was performed with 5 mL of 50 % ethanol solution to 0.5 g of the ground seeds, then it was sonicated by ultrasonic bath for 30 min, centrifuged and the supernatant was filtered by membrane filter in which 20 µL were injected on a liquid chromatography system coupled with diode array and ionization mass spectrometry detectors (LC-MS) equipped with a C18 column (250 x 4.6 mm, 5 µm, NST), as mobile phase was used water and methanol with 0.1 % formic acid, the flow rate was at 1.2 mL min-1. UV detection was at 372 nm. The phenolic profile was performed using the molecular weight and the mass fragmentation. In purple pitanga seeds (53.08 % of moisture) were identified flavonoids compounds as well as anthocyanidin as ions at m/z 433 indicating ellagic acid pentoside, quercetin 3-O-pentoside and pelargonidin 3-O-glucoside, kaempferol-O-pentoside (m/z 417), cyanidin 3-O-glucoside (m/z 447) and quercetin (m/z 301); In red pitanga seeds (56.90 % of moisture) were identified anthocyanidin as cyanidin 3-O-glucosid (m/z 449) and flavonoids as myricetin 3-O-hexoside (m/z 479), myricetin 3-O-pentoside (m/z 449) and also ellagitannins as HHDP hexoside (m/z 481). Both varieties presented mostly quercetin derived and delphinidin compounds. Results showed important phytochemicals of which flavanols were the most predominant chemical group, evidencing interesting bioactive activity that may turn out enrichment of other products.
