IDENTIFICATION AND QUANTIFICATION OF THE MAIN NON-VOLATILE COMPOUND OF PURPLE PITANGA (Eugenia uniflora L.) PULP

The Brazilian cherry or pitanga tree is a plant that produces fleshy fruits and is sources of functional compounds, rich in vitamins and in compounds with antioxidant activity. This study aimed to identify and quantify the main compound from the non-volatile fraction of this fruit, evaluating different solvents and injection times. It was identified that the major compound in purple pitanga pulp is cyanidin-3-glucoside (C3G). To determine the most efficient solvent it was performed a dilution of the pure C3G pure in different solvents as described below: acetone:water (70:30), acetone:water:acetic acid (70:29:1) and acetone:water:hydrochloric acid (70:20:10), followed by the mixture of the solution in tubes stirrer for 2 minutes and centrifugation (4500 x g/4°C /1 min.) The samples were injected at 0, 30 and 60 minutes after dilution into each of three solutions. For quantification of C3G in purple pitanga, the separation of the non-volatile fraction was performed using the Flavour Solvent Assisted Evaporation (SAFE) technique in 100 mL of pulp. At 30 and 60 minutes after the dilution C3G in acetone:water:hydrochloric acid (70:20:10) approximately 18 and 26%, respectively, of C3G had been degraded, while in the solution of acetone:water:acetic acid (70:29:1) only 2 and 6% were degraded, respectively. Using these results, the extraction of C3G using a solution of acetone:water:acetic acid (70:29:1) extraction under the same conditions described in test was chosen. A standard curve was prepared with the pure C3G and through this curve, it was observed that the pulp has cherry 340 µg/mL of C3G.
