CAROTENOIDS ENRICHED EXTRACT FROM CASHEW BYPRODUCTS – FIRST TOXICOLOGICAL EVALUATION

Many synthetic dyes, widely used in food and pharmaceutical industries, were associated with health concerns such as carcinogenesis. EMBRAPA/Fortaleza in association with CIRAD/France has developed an industrial process to obtain a carotenoid-enriched extract (CEE) from cashew apple fibrous residues. The CEE showed an intense yellow color with 11 carotenoids already identified pointing out to the CEE potential as a natural dye with interesting nutritional value. Thus, the present study aimed the initial CEE safety evaluation. The Embrapa Tropical Agroindustry prepared CEE using Embrapa CCP-76 cashew peduncle grown in the experimental field (Pacajus - CE, Brazil). The CEUA/UNICAMP approved the in vivo acute oral toxicity assay (protocol no. 4345-1). After 12h fast, female Unib:SW adult mice (25-30 g body weight, n = 5 animals/group) were randomly distributed in to experimental groups (G 1 - 4) which were orally treated with decreasing doses (1000, 500 and 250 mg/Kg) of CEE or vehicle (PBS buffer with 2% of Tween 80, 10 mL/Kg, control group). General toxicity clinical parameters were observed continuously during 4h after treatment and daily during 14 days. Body weights were registered on 1st, 7th and 14th experimental days. On day 14, after euthanize, a macroscopic evaluation of internal organs were realized and organ weight were registered for liver, spleen and kidneys. Statistical analysis was performed using ANOVA followed by Dunnett test (p < 0.05 for significance). No change in body weights nor any clinical signs of toxicity were observed between the experimental groups and the control one during the experiment. None abnormality or organ damage were observed during macroscopic evaluation besides neither alterations in relative organ weight. Thereby, the CEE from cashew apple fibrous residues showed none acute oral toxicity at a dose level of 500mg/Kg, being this dose assumed as the maximum dose tolerable for further in vivo evaluations.
