EVALUATION OF THE CHEMICAL COMPOSITION, TOXICOLOGICAL AND ANTIOXIDANT POTENTIALS OF THE ESSENTIAL OILS OF CHILEAN BOLDO COMMERCIAL SAMPLES
Chilean boldo (Peumus boldus Molina) products are used to treat digestive and hepatobiliary disorders. Some of their pharmacological properties (e.g.: antioxidant action) are associated with the chemical composition of its essential oils, which also can have toxic compounds. Thus, the aim of this study was to characterize the chemical composition, toxicological and antioxidant potentials of the essential oils of Chilean boldo commercial samples packed on sachets [group 1 (G1): five samples] or plastic bags [group 2 (G2): four samples]. These oils were extracted by hydrodistillation (100ºC, 1 hour) and analyzed by GC/MS and GC/FID. The toxicological evaluation was carried out by Artemia salina bioassay. Six methanol solutions (5, 10, 30, 60, 100, 150 ppm) of each essential oil were tested. The antioxidant activity was evaluated by DPPH assay, using four methanol solutions of each essential oil (1.0, 4.0, 8.0, 12.0 mg/mL). Fifty-five compounds have been identified. Ascaridole [(28.12 ± 12.37)%], 1,8-cineole [(13.73 ± 2.95)%] and p-cymene [(11.07 ± 1.77)%] were the major ones in both groups. The mean IC50 value was (12.75 ± 1.24) mg/mL, being bigger than that found for rutin (IC50 = 0.093 mg/mL) and gallic acid (IC50 = 0.0085 mg/mL), both used as positive controls. No statistical differences (p>0.05) were noted between the IC50 values of G1 and G2 samples. The mean lethal dose value (LD50) of G1 [(24.73 ± 4.78) ppm] was not statistically different from that of G2 [(42.18 ± 28.75) ppm]. The similarity between the chemical profiles of G1 and G2 may indicate a single source for these products. The presence of the chemical markers p-cymene, 1,8-cineole and ascaridole suggests that the samples are genuine. All LD50 values were below 200 ppm, indicating that these essential oils were highly cytotoxic. Finally, they were weaker antioxidants than rutin and gallic acid.
