The Brazilian Cerrado biome is a source of plants with potential for the development of new drugs. Araticum (Annona crassiflora Mart.) has a unique chemical composition, rich in tannins, flavonoids and other bioactive compounds. Several parts of this plant have been used in folk medicine. However, their properties are still scientifically under-explored. This study aimed to characterize the crude extracts of araticum peel and seed, and evaluate their effects on migration of human epithelial cells. Milled freeze-dried seeds and peels (1g each) were extracted by sonication using methanol/acetone/deionized water (7:7:6, v/v/v; 3x; 1:15 w/v) affording seed (ASE) and peel (APE) extracts after solvent elimination. Chemical characterization of ASE and APE was carried out by UHPLC-ESI-(-)-MS/MS. Cell migration assay was performed in immortalized human keratinocytes (HaCat) treated with medium without any fetal bovine serum (FSB, negative control), 5% FBS enriched medium (positive control), ASE (1.8 and 3.6 μg/mL) or APE (18 and 36 μg/mL) during 24h after scratch. The cells were photographed at 0, 9, 18 and 24h, and images were analyzed by ImageJ. In APE, ten phenolic compounds were identified, being epicatechin (6221.63 μg/g) and catechin (579.40 μg/g) the major compounds, whereas in ASE quercetin (21.11 μg/g) and chlorogenic acid (16.41 μg/g) were the major constituents besides more ten phenolic compounds. After 24h, ASE promoted a wound closure of 73.3% (3.6 μg/mL) and 54.1% (1.8 μg/mL), while APE showed only 3.8% and 0.6% of wound closure for 36 and 18 μg/mL, respectively, in comparison to time 0h. The results demonstrate that araticum seed extract was a promisor wound healing agent, while araticum peel extract had promisor effect anti-migration that could be useful in the keloids treatment or even as adjuvant in cancer chemotherapy.
