Resumo

Yarrowia lipolytica-a non-conventional yeast-is recognized as GRAS (Generally Recognized as Safe), and it can be applied in several industries, such as food, cosmetics, and pharmaceutical. Moreover, it can be an economical source for extracting valuable intracellular bioproducts such as proteins, peptides, β-glucan, and surfactants. However, cells must be disrupted to release those intracellular yeast compounds. In this study, Y. lipolytica IMUFRJ 50682 cells were disrupted by two step-reaction: first autolysis (120 rpm, 60 °C, pH 6, 24 h, and 15% w/v), then acid hydrolysis (120 rpm, 60 °C, pH 2, and 60 min). The cell rupture rate and the total carbohydrates and proteins released after extraction were evaluated. The cell rupture rate was 55.6±2.5%, showing an excellent method to obtain intracellular bioproducts. The total carbohydrates released was 11.1±0.1g/L, and the total protein was 134.3±1.0mg/L. These results indicated that the combined extraction methods are promising to obtain intracellular bioproducts suitable for application in food and cosmetics.
