Ionic liquids increased the proteolytic activity of a commercial enzyme from Aspergillus oryzae

Resumo

Three ionic liquids (ILs) were tested to study the ability to change the structure of one protease and the improvement on its biochemical properties.). For this, the enzymes solutions were pretreated with the ILs Choline Acetate, Choline Lactate and Choline Hydroxide at 5, 10, 15, 20 and 25 mmol L-1.The results showed that the ILs significantly increased the proteolytic activity of the protease studied (Flavourzyme). The enzyme activity was proportionally affected by Choline Hydroxide concentration. This positive correlation was also detected for Choline Acetate and Choline Lactate at concentrations up to 15 mmol L-1. The gain of activity can be attributed to the increase in hydrophilicity of the substrate and improvement of the nucleophilicity of water in the vicinity of the enzyme through hydrogen bonding. In summary, the results obtained in this study showed that the treatment of proteases with ILs can be a valuable strategy to increase their enzyme activities
