Genome-wide association between single nucleotide polymorphisms with beef fatty acid profile in Nellore cattle using the Bayes Cπ method

This study aimed to identify genomic regions associated with fatty acid (FA) composition on Longissimus thoracis (LT) muscle from 963 Nellore bulls. The beef fatty acids analyzed were: sum of saturated fatty acids (SFA), sum of monounsaturated fatty acids (MUFA) and sum of polyunsaturated fatty acids (PUFA). The animals belonged to eight different regions of Brazil, which participate in the PAINT, Conex?o DeltaG Norte and Qualitas beef cattle breeding programs. A total of 963 Nellore bulls with an average age of 24 months and feedlot finished for at least 90 days were used. During the boning, samples of LT muscle were collected, for FA composition, being quantified by gas chromatography (GC-2010 Plus - Shimadzu AOC 20i auto-injector) with a SP-2560 capillary column (100m x 0.25mm in diameter with 0.02mm thickness, Supelco, Bellefonte, PA). Animals were genotyped using a panel with 777,962 SNP (High-DensityBovineBeadChip). The BayesC model with ? = 0.01%, was used to identify genomic regions associated with FA composition, where ? is the SNPs proportion different from zero. The gs3 software was used to perform the analyses. The model utilized included the fixed effects of contemporary group (farm-year-management group), animal age at slaughter (linear and quadratic effect), and the SNPs random effects. A total of seven SNPs explaining more than 1.0% of the genetic variance associated with SFA, 358 SNPs with MUFA and none with PUFA were identified. After submitted the candidate genes to the DAVID bioinformatics database, the following pathways associated with fatty acid were identified: oxidative phosphorylation (NMRAL1 for MUFA, and ATP6V0E1 for SFA), insulin signaling (CBL for SFA), lipid metabolism (GLYR1 for MUFA), carbohydrate transport and metabolism (GMS, TREH for SFA), pentose phosphate pathway (PGD for MUFA). This study provides a valuable insight into the biology of beef FA composition in zebu cattle under tropical conditions.
