Log inINTRODUCTION AND OBJETIVES: The extreme environment of the sea gave rise to the most extraordinary biodiversity on the planet. Marine biotechnology aims to exploit the potential of these still unknown resources for the development of new products and the discovery of solutions. In oncology, marine drugs have been shown to be potent antineoplastic agents, but none have yet focused on skin cancer, the most common all over the world and corresponding to a third of diagnoses in Brazil. In this study we investigate the anticancer, immunomodulatory profile and healing potential of an isolated compound from the red algae (Rhodophyceae phylum) and its association with nanoparticles in melanoma and non-melanoma cells.
MATERIAL AND METHODS: A secondary metabolite halogenated will be associated with natural oil based nanoformulations in increasing concentrations. The mechanisms of action will be tested in the cell lines of murine melanoma (B16F10), non-melanoma of murine (4T1) and human (A431), fibroblasts and keratocytes in 3D cell cultures. The cell viability assay will be performed by acidic phosphatase activity measured in spectrophotometry to determine the inhibitory concentration in 50% of cells. After, the anti-metastatic/healing effect of nanoformulation will be observed as the spheroid’s migration on gelatine. The cell death and proliferation rates will be assessed by immunofluorescence for activated caspase 3 and 9, and Ki-67 expression. The immunomodulatory cytokines profile will be evaluated by qPCR and ELISA.
RESULTS AND CONCLUSIONS: It is expected to demonstrate the in vitro mechanisms of action of natural bioactive agents through the mimetic tumor model and thus contribute for the development of the bioproduct nanotechnology, resulting in a novel biopharmaceutical of topic use for skin cancer treatment.

Olá! Sim, as nanoformulações com o produto natural passaram por ensaios citotóxicos em cultura de células 2D. No entanto, a nanoformulação final contendo o metabólito secundário da alga marinha ainda não foi testada devido à atrasos nos experimentos por conta da pandemia mas ela passará por ensaios de viabilidade e morte celular, migração e cicatrizante.

Na verdade, me refiro em relação a estabilidade dessas formulações.. fizeram algum teste? Ou não são necessários?

Sim. A estabilidade da nanoformulação sem o produto da alga já foi testada e aprovada pela ANVISA. No momento, estamos na fase de analisar a estabilidade da nanoformulação com o metabólito da alga marinha.
