The development of biotechnological processes involving the use of immobilized enzymes has been a significant highlight because advantages as stability and the possibility of reuse in continuous processes. In this work, lipolytic enzymes such as Thermomyces lanuginosus (TL), Candida antarctica B (CALB) lipases were immobilized by covalent binding by diversified process called “load” and “overload” in sol-gel matrices by diversified methodologies. In order to compare load and overload biocatalysts, commercial biocatalysts (TL IM and Novozyme 435) and the free lipases, the catalytic activities were measured on reactions of hydrolysis of olive oil and esterification of lauric acid with ethanol. TL and CALB lipases when immobilized by overload methodology showed 40 and 84 % of increase on the esterification activity, respectivelly. Also, the effectiveness of the immobilization by load and overload was confirmed by FTIR.
