Desenvolvimento de Plataforma Nanoestruturada para Teste Rápido de Hidronefrose
Hydronephrosis is the most common fetal urinary tract alteration, able to permanently damage the kidney when not precociously diagnosed[1]. Development of a rapid test for renal function evaluation related to this condition using cystatin C (cys C) is extremely important for newborns. Thus, a nanostructured platform for cys C detection without redox probe was developed. Amineferrocene (AFC) is a mediator with inherent redox activity[2], and can be used to functionalize nanomaterials in order to surpass this obstacle. Association with graphene oxide (GO) allows for an easy and cheap sensing platform. Herein, three solutions containing 1) aminoferrocene (AFC), 2) graphene oxide (GO) and 3) GO with AFC were prepared by ultrasonic bath for 1h and tested to observe the functionalization of GO with AFC. Concentration tests with AFC were also performed. After, anti-cystatin C was immobilized onto the surface (incubated for 1h) and free sites blocked with glycine to avoid nonspecific bonds. Functionalization of AFC/GO was observed by cyclic and square wave voltammetry (CV and SWV, respectively), obtaining results in KCl solution (0.1M). Following steps (immobilization and blockage) were also followed in KCl by SWV. The designed platform proved stable when used without redox probe. The antibody was successfully immobilized on the platform surface and nonspecific sites blocked with glycine, as observed in figure 1. Thus, the platform developed herein may enable fast and reliable evaluation of renal function immediately after birth, reducing initial treatment time and the consequent damage that delay may cause.
