DIRECT qNMR MEASUREMENTS

Abstract

Nuclear Magnetic Resonance spectroscopy can be considered a primary method of measurement as the relation of substances can be determined directly from the physical context of the measurement without referring to another compound. [1] Additionally, NMR experiments are intrinsically quantitative as there is a direct ratio between the strength of the response signal and the number of nuclei responsible for that signal. Therefore, quantification can be performed simply by obtaining a signal’s integral. The major advantage of qNMR over other spectroscopic methods is that in general not much sample preparation is required. The mathematical theory that involves these facts is widely disseminated in the literature.[2]

Although the topic is widely addressed in the literature, exploring it experimentally has been a challenge. Different qNMR methods have been proposed, usually depending directly or indirectly on some reference material. However, qRMN quantifications can be made directly, as the absolute integral of the signals of interest and the molar concentrations of the analyte in question are directly correlated. As this quantitative determination is independent of the studied molecule, there is no need to use internal or external reference material.

Dear Lorena, thank you for your interest.
Sorry for the bogus presentation. In fact, the integrals vary with the concentration of reference material, as we expected. But on the slide it just gives you the weights, instead of the concentration. You can see from the superposition of the different linearity graphs in the end that actually the signal intensity actually is independent of the molecule involved, which is what we want to explore better in the future.
Cheer, Jochen

Ah ok! Thanks for the answer Jochin! Good to ser you again even in virtual form. Cheers
