Oxindole alkaloids biosynthesis in Uncaria tomentosa (cat’s claw)

Abstract

The specie Uncaria tomentosa is used for medicinal purposes dating back over 2000 years1 and widely used worldwide to complement therapies for treating inflammatory diseases and cancer1, in addition to its anti-oxidant, antidiabetic, antimicrobial potential and against the effects of Parkinson disease2. Oxindole alkaloids are biosynthesized by cat’s claw and are the compounds responsible for pharmacological activities. Having established a substrate feeding protocol for Cat’s claw plantlets, the biosynthetic route for mitraphylline (1), isomitraphylline (2), pteropodine (3) and isopteropodine (4) was investigated using 2-13C-tryptophan, 3-13C-sodium pyruvate, and 1-13C-DL-glyceraldehyde. The biosynthetic pathway of 1-4 are derived from the MEP and shikimate pathways. This experiments also allowed us to establish the biosynthetic pathway for oxindole alkaloids from U. tomentosa with 13C-precursors for the first time in the literature.
