Galenical development of orodispersible films formulations for incorporation of hydrophilic drugs

Abstract

Alternative dosage forms have been prepared in compounding pharmacies to fill a gap created by the lack of individualized products. In this scenario, orodispersible films (ODFs) appear as a promising alternative. The aim of this work was to develop polymerics solutions to prepare ODFs containing hydrophilic drugs. Different combinations of polymers and others excipients were tested. The casting-solvent evaporation technique was used to prepare the films: the solutions were casted onto a glass surface using an applicator for preparative thin layer chromatography plates (Figure 1). The quality parameters evaluated in films were appearance, thickness, weight variation and disintegration time. The solutions containing pullulan, xanthan gum, carrageenan or gelatin were considered most suitable for casting. All formulations yielded smooth films on both sides; no lumps and insoluble solids; bright; transparent; and resistant to handling. Gelatin films showed the shortest disintegration times in water and in simulated saliva buffer (< 60 s).
