5-HT1A receptor participation in anxiolytic-like and antidepressant-like effects of compound 2-(4-((1-phenyl-1H-pyrazol-4-yl)methyl)piperazin-1-yl ethyl acetate (LQFM192)

Abstract

LQFM192 is a piperazine derivative that has demonstrated anxiolytic-like and antidepressant-like activities in elevated plus maze (EPM) and forced swimming (FS) tests, respectively. In this sense, objective of this study is to investigate the action mechanisms involved in these activities. For this, Swiss male mice (n=8) were pre-treated (i.p.) with 0.9% saline or WAY-100635 (0.7 µmol/kg) and after 1 hour were treated (p.o.) with vehicle (10 mL/kg) or LQFM192 (54 µmol/kg). Sixty minutes after the treatments, mice were submitted to EPM and FS. LQFM192 treatment compared to vehicle increased percentage of time spent in open arms of 42.55 ± 1.52 to 52.69 ± 2.38 in EPM and decreased the time of immobility of 193.67 ± 7.88s to 99.14 ± 12.09s in FS. However, anxiolytic-like and antidepressant-like effects were reversed by pre-treatment with WAY-100635 in 40% and 33%, respectively. Together, the results suggest involvement of 5-HT1A receptor in both activities.
