Coumarins are oxygenated heterocyclic compounds widely distributed in nature, mainly in plants. Their diverse
pharmacological properties have motivated medicinal synthetic chemists to synthesize new structures aimed
at treating various diseases. Therefore, there are currently several reactions for obtaining these heterocycles,
such as Pechmann, Wittig, Knoevenagel, or Perkin.1 In previous works, we published the synthesis of
coumarins through Pechmann reaction and obtained amine compounds with in vitro activity against
Trypanosoma cruzi and, combining the Pechmann and Huisgen reactions, coumarin/1,2,3-triazole hybrid
compounds with marked antitumor activity against breast cancer cell lines (MCF7)2,3. Isoxazoles are
heterocycles widely used as fragments in the structures of drugs or pesticides.4 Therefore, continuing our
studies, in this work, we present the synthesis of novel coumarin-isoxazole hybrids. Coumarins derivatives 2 and 3 were
synthesized through Pechmann R. with phenols 1 and -ketoesters a and b, and furnish pure compounds
(according NMR spectra) in moderate to good yields (38%-59%).
