The emergence of drug-resistant Plasmodium falciparum strains and limited therapeutic options for leishmaniasis underscore a global health demand for novel anti-parasitic agents [1]. To combat this challenge, a study successfully designed, synthesized, and biologically evaluated an innovative series of fifteen hybrid phthalimide-linked piperidine derivatives specifically targeting P. falciparum heat shock protein 90 (PfHsp90) [2]. The research utilized an efficient acetylenic Mannich reaction with copper iodide as a catalyst to build a diverse compound library. Evaluations included structural characterization, modeling, ADMET predictions, and biological testing. Compounds Am4 and Bm4, displaying strong PfHsp90 binding affinity, moderate potent in vitro antiplasmodial activity, favorable pharmacokinetic properties, and low cytotoxicity profiles [3]. Additionally, compounds Am6 and Bm6 exhibited potent in vitro activity against Leishmania infantum, with IC₅₀ values of 2.80/2.73 and 2.02/2.12 µM at 24/72 h, respectively, and favorable 24-h selectivity indices of 49.7 and 43.9, highlighting their exceptional potential as antileishmanial lead compounds.
