Midazolam (I) is a benzodiazepine derivative widely used as a sedative, anxiolytic, and anesthetic induction agent.1 Its synthesis requires an oxidative aromatization step to generate the bioactive pharmaceutical ingredient.1-3 In this work, a continuous-flow process for the oxidative aromatization of the non-aromatic precursor of midazolam (II) was developed using a packed-bed reactor containing MnO₂ as a heterogeneous oxidizing agent.4 Process optimization was performed through a design of experiments (DoE), evaluating substrate concentration, temperature, and MnO₂ mass at a fixed residence time of 30 min. Conversion was determined by HPLC-PDA. All variables significantly affected the process, with temperature identified as the most influential factor. Under optimized conditions (0.017 g mL⁻¹ substrate concentration, 110 °C, and 6 g MnO₂), conversions above 99% were achieved within 30 min. In comparison, the analogous batch process reached 97% conversion only after 12 h, highlighting the superior productivity and efficiency of the continuous approach.
