Gold Nanoparticle-Based Dynamic Light Scattering (DLS) Immunosensor for Zika Vírus

Abstract

This work describes the development of a gold nanoparticle (AuNP)-based immunosensor for the detection of Zika virus (ZIKV) E2 protein using Dynamic Light Scattering (DLS). DLS analysis revealed a progressive increase in hydrodynamic diameter (DH) from 49.51 to 89.21 nm after polyclonal antibody (pAb) conjugation. Incubation with ZIKV E2 protein (0–50 µg/mL) demonstrated that the immunosensor maintained its colloidal stability throughout the evaluated concentration range. In contrast, Dengue virus (DENV) and Chikungunya virus (CHIKV) proteins, each evaluated at 8.33 µg/mL, induced increases in DH of up to 30%, confirming the platform's high analytical selectivity for arbovirus differentiation. Overall, this platform represents a promising strategy for the rapid screening of viral biomarkers.
