ALTERNATIVE MEDIA FOR HYALURONIC ACID PRODUCTION BY Streptococcus zooepidemicus ATCC 39920
The search for sustainable socio-economical development has stimulated studies using alternative media for fermentation processes. The sugar cane molasses, sugar cane industry by-product, has been used at the production of interest metabolites for the reason of its high concentration of sucrose, glucose, fructose, salts, and some sources of nitrogen, essential to microbial metabolism. Considering that 80% of hyaluronic acid production costs are due to nutrients required for the Streptococcus growth, this research aimed at assessing the polymer production in media containing non-clarified and clarified sugar cane molasses. The molasses was obtained from Altech group (São Pedro do Ivaí, PR, Brazil), centrifuged at 9000 rpm, 4°C, 15 min and, in order to obtain clarified molasses, the supernatant was treated with 8% w/v of activated charcoal, at 70°C for 1 h, and centrifuged at 9000 rpm, 4°C for 15 min. The fermentations were performed in 125 mL Erlenmeyers with 25 mL of media (g.L-1): non-clarified or clarified molasses, 30 total sugar; yeast extract, 30; K2HPO4, 2.5; NaCl, 2.0 and MgSO4, 1.5. The inoculum was 0.2 g.L-1 and the assays were carried out at 100 rpm, 37°C and pH 8.0 for 24 h. Hyaluronic acid, lactate, formate, acetate and ethanol were evaluated in high performance liquid chromatography (HPLC), with refractive index detector. For the hyaluronic acid OHpak-SB-806M HQ 80 x 300 mm column was used, at 40°C, and NaNO3 0.1 M at 1 mL.min-1 as mobile phase. Lactate, formate, acetate and ethanol were studied by means of Aminex HPX-87H 300 x 7.8 mm column, at 60°C, and H2SO4 0.005 mol.L-1 at 0.7 mL.min-1 as mobile phase. The media was compared by Tukey test at 5%, using the Software Statistica 9.0. The greatest hyaluronic acid production was 0.662±0.003 g.L-1 in clarified molasses and this was statistically different from the production obtained in non-clarified molasses, 0.557±0.038 g.L-1. The synthesis of acetate differed from the non-clarified molasses, 0.582±0.031, and clarified, 0.678±0.018 g.L-1. The lactate production was 4.021±0.229 and 4.315 ±0.161 g.L-1 in non-clarified and clarified molasses, respectively, not statistically different. Formate and ethanol were not detected.
