Abstract

Introduction and objectives: DNA is constantly assaulted by several endogenous and exogenous factors in the cell. Mechanisms for maintenance of DNA integrity that recognize and repair damages have evolved over the course of evolution. Deficiencies in DNA repair pathways contribute to an accumulation of mutations that may result in cancer. The double strand break (DSB) is one of the most toxic lesions that affect DNA and there are two major pathways of repair: the non-homologous end joing (NHEJ) and the homologous recombination (HR). The HR is known as an error free repair pathway - it uses the sister chromatid as template for to resolve the damage. PALB2 is one of the key proteins in HR, working as a linker between BRCA1 and BRCA2 and on the recruitment of the RAD51 recombinase to DNA damaged site. Further, germline mutations in the PALB2 gene have been implicated in breast and pancreas cancer susceptibility. But despite the growing interest in PALB2, little is known about its promoter region. In this work, our goal is to characterize the PALB2 promoter locus and look for regulatory elements to better understand its expression control. Materials and methods: The putative PALB2 promoter region was determined by in silico analysis, predicted transcriptional factors were identified using the PROMO platform. Among others, regulatory elements associated to the tumor suppressor TP53 were found. The putative promoter region (1001 nucleotides), deletions fragments enclosing the last 366 or 142 nucleotides at the 3’ end and also a fragment containing the first 635 nucleotides at the 5’ end were generated through PCR using BJ cells genomic DNA as temple. Fragments were cloned into pGL3-basic vector. Promoter activity was assessed in the HEK293FT cells through a reporter expression assay after DNA damage induced by ionizing radiation (IR) or not. We also investigated the contribution of wild-type TP53 in the regulation of PALB2 by analyzing PALB2 protein levels in human cells TE-1 by immunoblotting. The cell line expresses a temperature sensitive conditional TP53 mutant: dysfunctional at 37ºC but wild type like at 32°C. Results and conclusion: Preliminary data indicate that 1001 and 366 fragments present a similar promoter activity (eight times higher than the negative control, empty vector). However, the 635 and 142 fragments presented no promoter activity at all, similar to the negative control. The 1001 and 366 fragments also showed an increase in promoter activity when cells were treated with IR (10Gy), suggesting that PALB2 is upregulated in damaged cells. Interestingly, TE-1 cells showed higher PALB2 protein levels when cultured at 32°C than at 37°C when TP53 is dysfunctional, the phenotype is increased in irradiated cells. These data suggest that the last 366 nucleotides at the 3’ end in the PALB2 putative promoter region encloses a minimal regulatory region for PALB2 expression and the TP53 protein may contribute to PALB2 regulation.

Boa tarde, Dario. Para os ensaios de luciferase foram utilizadas as linhagens HEK293FT, HeLa  e HCT116, e os resultados obtidos foram muito semelhantes em todas as linhagens testadas.

Já em relação ao western blottings para avaliar a influência de TP53 nos níveis proteicos de PALB2, utilizamos a HEK293FT para super expressar p53 (observamos um aumento nos níveis proteicos de PALB2 em relação às não super expressas), na linhagem termo sensível TE-1 (onde observa-se maior nível de PALB2 quando eu tenho p53 funcional - à 32ºc) e em HCT116 wt e HCT116 p53-/-, onde também foi possível observar um maior nível de PALB2 nas células selvagens em relação às silenciadas para p53. Com isso, a gente pode dizer que os dados sugerem sim, uma interação de p53 na região promotora de PALB2 atuando como um dos fatores de transcrição responsáveis pela regulação do seu gene.

Muito obrigado pelas suas respostas esclarecedoras. Mas ainda tenho duas duvidas:

1) Tu tentaste realizar (ou pretende realizar) experimentos de marcacao com imunohistoquimica ou mesmo de citometria de fluxo para avaliar o estudo de possiveis faores de regulacao de PALB2 ("Possible regulatories factors of PALB2 promoter:") ao inves de WESTERN BLOT, o qual pode apresentar algumas disvantagens como falso-positivos?

2) Na Figura 2A a qual mostra a analise "in silico" de possiveis fatores que se interagem a regiao promotora de PALB2 ("All transcriptional factors predicted by AlygnPROMO that may bind to this region"), foi selecionado o p53. Tu tambem pensaste em selecionar outro fator indicado pela analise, mesmo com "baixa probabilidade",  como por exemplo c-Fos ou mesmo PEA3, talvez como uma especie de "controle"?

Então:

1) Os próximos experimentos que pretendemos fazer para analisarmos os possíveis fatores de transcrição de PALB2 são:

-  Principalmente, uma vez caracterizada a região mínima necessária para a atividade promotora de PALB2, pretendemos biotinilar essa região para conseguirmos isolá-la e realizar uma espectometria de massas para identificar as proteínas que estão interagindo com ela

- Fazer uma imunopreciptação de cromatina para confirmar a presença dos possíveis fatores de transcrição no promotor de PALB2.

- Super expressar e talvez silenciar o fator de transcrição alvo de análise e realizar o ensaio repórter de luciferase para ideinftificar se houve um aumento ou diminuição na atividade promotora do promotor de PALB2.

2) Com certeza a gente pensa em analisar outros fatores de transcrição presentes na lista, sim. O TP53 foi nosso primeiro devido a disponibilidade de ferramentas no laboratório, mas pretendemos explorar melhor essa lista. Porém, primeiro gostaríamos de caracterizar melhor a região promotora e identificar a porção mínima necessária para essa atividade, o que nos facilitará a identificar os possíveis fatores de transcrição que se associam à ela.

Olá, Fernanda, muito obrigada! 

Nós selecionamos inicialmente o TP53 por ser uma proteína que já possui um papel de resposta a danos no DNA muito bem caracterizado e devido a disponibilidade de ferramentas que temos no laboratório. Outros experimentos ainda serão realizados para ajudar na caracterização dessa possível regulação de PALB2 através de p53, como por exemplo um ensaio de luciferase de células com a super expressão ou silenciamento de p53 ou até mesmo uma imunopreciptação de cromatina. Mas, com certeza, pretendemos estudar e analisar outros putativos fatores de transcrição para entendermos como funciona a regulação de PALB2.

Porém primeiro a gente gostaria de melhor caracterizar e mapear a região promotora de PALB2 identificando a região mínima necessária para a atividade promotora. O que nos facilitará entender melhor como funciona a regulação dessa proteína, além de ajudar na identificação dos possíveis fatores de transcrição responsáveis pela sua regulação.
