INTRODUCTION AND OBJECTIVES: Melanoma is a very common and aggressive malignancy that arises within the skin and affects millions of people in the world. The skin microenvironment is complex and highly regulated by immune cells and their crosstalk with different nerve types. Sensory nerves are present in the skin, where they control immune cells during different processes in the skin (i.e. infection and growth disorders). Moreover, we have identified, in mouse models, that these nerves are able to infiltrate the melanoma microenvironment and their ablation by genetic and pharmacological approaches favors tumor growth. These findings suggest that sensory nerves interact with components of the melanoma microenvironment, controlling its growth. Since regulatory T cells (Tregs) and Myeloid Derived Suppressor Cells (MDSC) are present within the tumor microenvironment and regulate antitumor response, we sought to evaluate these cells in the melanoma microenvironment of sensory nerve-ablated mice. MATERIAL AND METHODS: To perform sensory nerve ablation, 4-week-old FoxP3-GFP mice were injected subcutaneously on consecutive days with increasing doses of RTX (30, 70 and 100 μg/kg), control mice received vehicle alone. After a resting period of 20 days, mice were subcutaneously injected with 1x105 B16F10 cells, followed up for 20 days and euthanized. Tumors were dissected, weighted and processed for flow cytometry. After labeling the cell suspension with antibodies against surface markers of lymphoid cells and MDSCs, cells were analyzed on a FACS CantoII. RESULTS AND CONCLUSION: RTX-treated mice developed larger tumors, as determined by tumor weight and tumor weight/body weight. Flow cytometry analyses showed, in RTX-treated mice, a decreased frequency of CD4+ and Treg cells (not significant), accompanied by an increased frequency of CD4- cells. No significance was observed in the frequencies of MDSCs, macrophages, monocytes and neutrophils. Interestingly, when we analyzed the expression of the exhaustion marker PD1 in different lymphocyte (CD3+) populations we observed an increase in TregPD1+ cells and a decrease in CD4-PD1+ cells. In summary our results suggest that the ablation of sensory nerves lead to a regulatory profile of immune cells, which supports tumor growth. Nevertheless, further studies should be carried out to better understand the characteristics of these cells in the melanoma microenvironment.

Quais seriam os próximos passos experimentais a serem tomados com objetivo de entender melhor as caracteristicas dessa celulas no microambiente do melanoma? Se você tivesse que convencer por ex o Instituto Serrapilheira a financiar seu estudo o que proporia?

Temos algumas propostas no nosso grupo, uma vez que esses dados ainda são preliminares. (1) Outros marcadores devem ser utilizados para formar um painel mais completo e caracterizar melhor as células que estão em exaustão. (2) As células podem apresentar um perfil de marcadores que indica sua exaustão, mas devemos fazer experimentos funcionais para validar esses perfis. (3) Várias outras células do microambiente do melanoma podem participar da modulação da resposta imune tumoral, então devemos avaliar outras células dentro desse contexto para compreender melhor o processo.
Como propostas de experimentos: (1) Isolar por FACS diferentes populações de linfócitos CD8+ e CD4+ com base (principalmente) na expressão de PD-1, CTLA4 e CD25 e avaliar sua funcionalidade em cultura. (2) Avaliar populações de células tronco mesenquimais/estromais e células tronco tumorais no melanoma e correlacionar a expressão de marcadores dessas células com sua "conversa" com células imunes. (3) Avaliar como (e se) a ablação de nervos sensoriais ou sua modulação pontual pode melhorar a resposta a imunoterapias.
