Polygalacturonase production by thermophilic Bacillus sp SMIA-2 using passion fruit rind flour
Application of wastes from the food processing industry as carbon sources in enzymes production processes reduces the cost of production, and also helps in solving problems of their disposal. In this work was studied the utilization of passion fruit rind flour (PFRF) for polygalacturonase production by a thermophilic Bacillus sp. SMIA-2. Scanning electron microscopy studies showed that the organism attached firmly to the substrate and showed maximum polygalacturonase activity after 129 h of incubation at stationary phase of cell growth. The PFRF concentration used in the culture medium had a positive effect on growth of the organism but, had a negative effect on enzyme activity. Regarding to the initial pH medium, the values between 7.0-7.5 were the best for polygalacturonase secretion. Studies on enzyme characterization showed that the dialyzed polygalacturonase presented an optimum activity at 70 °C and pH value of 8.5.
