Selection of isolates of Antarctic psychophilic bacteria for peptidase production and carbon source impact assessment

Abstract

The aim of this work was to select the best producer of peptidases among bacterial isolates collected at different locations in Antarctic soil and evaluate the impact of different carbon sources on enzyme production. Cultivations were performed on a rotary shaker for 144 h, at 15 °C and 100 RPM, throughout which biomass formation, substrate consumption, production of peptidases and pH variation were evaluated. Isolates 11, 58 and 64 presented the highest peptidase activities, reaching 349.17, 335.58 and 365.42 U/L in 72 h of cultivation, respectively. Soil isolates associated with a rookery had higher peptidase activities compared to those from other areas. Biomass and enzyme activity values were lower in the medium with addition of glucose and higher when glycerol was added compared to the original LB medium. Within 72 h of cultivation, peptidase production increased approximately 33% for the 1% glycerol medium and 57% for the 5% glycerol medium.
