Evaluation of 5-HT1A receptor involvement in anxiolytic-like and antidepressant-like activity of (1-(4-(3,5-di-tert-butyl-4-hidroxybenzyl)piperazin-1-yl)ethan-1-one) - LQFM211

Abstract

LQFM211 is a piperazine derivative that has been shown in previous studies to have anxiolytic-like and antidepressant-like activity in the elevated plus maze (EPM) and forced swimming (FS) tests. Thus, the objective was to investigate the mechanism of action of this compound, for that swiss male mice (n=8) were pretreated (i.p) with 0.9% saline or WAY-100635 (0.7 µmol/kg) and after one hour were treated with vehicle (10 ml/kg) or LQFM211 (50 µmol/kg). Sixty minutes after the animals were submitted to EPM and FS. LQFM211 treatment compared to vehicle, increased percentage of time spent in open arms of 43.33 ± 3.37 to 54.10 ± 1.94 in EPM and decreased the time of immobility of 173.8 ± 13.41s to 124.9 ± 13.19s in FS. However, anxiolytic-like and antidepressant-like effects were reversed by pre-treatment with WAY-100635 in 17% and 63%, respectively. Together, the results suggest involvement of 5-HT1A receptor in both activities.
