Development and characterization of nanostructured lipid carriers for cannabidiol nasal administration

Abstract

Cannabidiol (CBD) is a lipophilic molecule with low aqueous solubility and an extensive first-time metabolism by the liver, making its oral bioavailability extremely low. In order to increase the CBD bioavailability and effectiveness, the aim of this study was to develop nanostructured lipid carriers loaded with CBD (CBD-NLC) for nasal administration. CBD-NLC were prepared by hot microemulsion technique using stearic acid, oleic acid, Tween 20 and cetyl pyridinium chloride. And they were characterized by size and zeta potential measurements, atomic force microscopy (AFM), differential scanning calorimetry (DSC), infrared spectroscopy (ATR-FTIR), and X-ray diffraction (PXRD). CBD-NLC with size below 180 nm, zeta potential of 41.55 mV, and high drug encapsulation were obtained. AFM analyzes showed spherical smooth surface particles. And DSC, ATR-FTIR, and PXRD studies suggested the incorporation of CBD in molecular dispersion into the lipid matrix. These results demonstrate the potential of NLC for nasal administration of cannabidiol.
